I don't believe this can be set using the native tools. It looks like this field is dynamically generated and listed on the quick forms for this panel. For example, the "App for Outlook Contact Card" doesn't have a lookup field on is for the "Set Regarding".
What if the user clicks on a related record towards the bottom of the panel? Such as a Case or Opportunity? This will open that record and the "Set Regarding" button is at the top. Thus, you are bypassing the lookup field you highlighted.

For a client I like to see if we can set the 'Set Regarding' field in the D365 App for Outlook to mandatory. They're not liking the fact that users can forget to set the regarding field which makes emails 'disappear' in the Dynamics black hole...
The only thing I can find in customizing the app, is the card that shows the first 3 fields of the entity.
