No, that's not how duplicate detection works. Duplicate detection works essentially two ways:
1. When creating a new record, if it trips a duplicate detection rule, it will pop a window showing the possible duplicate record and ask if you really want to create the new record.
2. A scheduled job that will provide a report of all potential duplicates based on your rules for a certain entity.
One you have identified the duplicates, the proper process is to merge the duplicate records and disable the duplicate so it's no longer used.
So for Advanced Find, the way to exclude duplicates is to go through the process of merging and disabling dupes, and then searching only for active records.

Not a unique list of all active contact email addresses, a list of active addresses excluding all addresses identified as duplicates.
I do have the OOB duplicate detection enabled on exact match on Contact email address, so is there something that exists on the Contact record identifying it as a duplicate that I could use?
